Figure 8. Differential effects of Zn2+ on biophysical properties of Kv3.1bHA and Kv3.1bHAC208A.

Figure 8

Kv3.1bHA and Kv3.1bHAC208A were transfected with YFP into HEK293 cells. Voltage-clamp recording was performed on these cells using the same protocol as in Fig. 2A. A, 100 μm Zn2+ inhibited Kv3.1bHA (top) but not Kv3.1bHAC208A (bottom). B, differential inhibitory effects of Zn2+ at low concentrations on current amplitude of Kv3.1bHA and Kv3.1bHAC208A. C, Zn2+ had no effect on the slope factor, k, in the G–V relationship of both Kv3.1bHA and Kv3.1bHAC208A. D, 30 μm Zn2+ significantly increased V1/2 of Kv3.1bHA but not Kv3.1bHAC208A. E, 30 μm Zn2+ slowed the activation of Kv3.1bHA but not Kv3.1bHAC208A at +30 mV. F, effects of Zn2+ at different concentrations on the activation time constant (τon) of Kv3.1bHA and Kv3.1bHAC208A. Paired t test: ***P < 0.001; **P < 0.01; *P < 0.05.
