Schematic representation of pTD-plasmid maps. Plasmid variants are constructed for periplasmic or cytoplasmic expression and different tag- and reporter gene combinations. Plasmid variants are equipped with the lacI^q-Ptrc promoter, the RK2 broad host range origin of replication (oriV/trfA) and the streptomycin/spectinomycin (aadA) or kanamycin (aphA) resistance gene (compare Table 1). Compatibility with SEVA enables exchange of variable modules like replication origin and antibiotic marker via the indicated restriction sites, while T0, T1 transcriptional terminators and the conjugation origin oriT are permanent elements.
