Please note that esiRNA–template production consists of two consecutive PCRs. In the table only the transcript-specific primers for the first PCR round are listed. Universal tags have to be added to all primers that allow amplification with universal T7 primers in the second PCR round. The universal tags are 5′-TGTAAAACGACGGCCAGT-3′ (forward) and 5′-AGGAAACAGCTATGACCAT-3′ (reverse). The universal T7 primers are 5′-GCTAATACGACTCACTATAGGGAGAG-3′ (forward) and 5′-GCTAATACGACTCACTATAGGGAGAC-3′ (reverse).
