UC-MSCs induce granulocytic differentiation of NB4 cells by activation of its intracellular MEK/ERK signaling pathway and IL-6 also functions through this pathway. (A) NB4 cells pretreated with PD98059, U0126, or SB203580 for 1 h were incubated with UC-MSCs or/and 10 nM ATRA and the CD11b level was detected by flow cytometry after 48 h culture. (B) Activation status of MEK1/2 and ERK1/2 in NB4 cells after having being treated by UC-MSCs or/and 10 nM ATRA for 24 and 48 h was illustrated by western blot. (C) NB4 cells pretreated with PD98059, U0126, or SB203580 for 1 h were stimulated by IL-6 (50 ng/mL) and the CD11b expression level was determined by flow cytometry after 48 h incubation.
