Strategy for the synthesis of lispro insulin, using an optimized chemical tether in the mini-proinsulin precursor 5: (i) oxime-forming ligation; (ii) folding/disulfide formation; (iii) enzymatic cleavage. Lispro insulin is a fast-acting analogue of native human insulin in which the ProB28-LysB29 sequence is inverted to LysB28-ProB29.19
