Small molecule inhibitors of DHHC protein acyl transferases. (A) 2-bromo-palmitate, cerulenin, and tunicamycin are reported generic inhibitors of palmitoylation(63–65). Electrophilic functional groups (red) facilitate irreversible alkylation. (B) The p-nitrophenyl benzylidine inhibitor, Compound V (CV)(68), absorbs visible light. The benzylidine Michael acceptor reacts with excess β-mercaptoethanol, breaking the conjugation and shifting the absorbance to the p-nitrophenyl chromophore (unpublished data). Experiments were performed at room temperature in phosphate buffered saline at the concentrations listed.
