(A) The codon-optimized ctxB gene was cloned upstream and in-frame with codon-optimized pfs25, which contains four tandem epidermal growth factor-like domain repeats. The resulting ctxB-pfs25 fusion gene was integrated into the psbA locus by homologous recombination such that transgene expression is driven by the light-regulated psbA promoter.
