Figure 1. — Map of the rearranged Ig light chain locus in the chicken B cell line DT40 ψV knockout. (Not to scale.) The locus contains a leader, VJ, and C region of the IgL gene. The strategy of knocking in the human β-globin transcription terminator (∼1.4 kb) by the targeted integration is shown. The terminator is knocked-in at +166 bp from the transcription start site, and 60 bp of DNA fragment containing part of leader – VJ intron and 8 bp of V region was deleted (from +166 to +225 from transcription start site).
