Purified N-ras+/+ OT-I CD8+ T cells were pretreated or not with the indicated inhibitors for ERK kinase (PD98059) and PI3K (wortmannin), stimulated with 10−9 M OVA peptide as in Fig. 7 D, and incubated in the presence of the inhibitors for the indicated times (A), or similarly stimulated and treated with the inhibitors for ERK kinase (UO126), PI3K (LY294002), PI3Kα (inhibitor VII) PI3Kβ (VI), PI3Kγ, or AKT (VIII) at graded concentrations, and stained for Eomes after 48 h (B).
