Suppressor T cell assays in 2-3 month old animals. Purified CD4+CD25+ regulatory T cells were cultured with CD4+CD25 responder T cells (at the indicated Treg:Tresp ratios) in the presence of beads coated with anti-CD3 and anti-CD28 mAb (for details see Materials and Methods). Inhibition of CD4+CD25 T cell proliferation relative to a 0:1 Treg:Tresp ratio was measured by [3H]thymidine incorporation. A) The suppressive function of Tregs from B6.129-Sle16Sle18 and B6.129–Sle16 mice was compared with that of B6 Treg cells. B) The sensitivity to suppression of responder T cells from B6.129-Sle16Sle18 and B6.129–Sle16 mice was compared with that of B6 Tresp. C) The suppressive activity of Tregs from B6.129–Sle16 or B6 mice was tested using Tresp from B6.129–Sle16 mice. D) The susceptibility to suppression by B6.129–Sle16 Tregs of Tresp from B6.129–Sle16 or B6 mice was compared. Results shown (mean ± SD) are representative of three independent experiments and were performed on 2-3 month old mice.
