Pygo2 binding facilitates the association of β-catenin and a bivalent chromatin configuration at the Notch3 locus in MaSC/basal cells. In Pygo2-null MaSC/basal cells, Notch3 chromatin shifts precociously from having dominant repressive H3K27me3 marks to prominent permissive H3K4me3 marks, allowing derepression/activation of Notch3.
