(a-c) Extensor digitorum longus (EDL) muscles were dissected and incubated in the presence or absence of ER stressors, tunicamycin (1 μg/ml) and thapsigargin (1.3 μg/ml), for 4 hrs. After the incubation, EDL muscles were collected and used for mRNA measurement for TRB3 (a) (n=5). Insulin-stimulated glucose uptake from EDL with tunicamycin (b) and thapsigargin (c) was determined as described in Experimental procedure (n=5). (d,e) EDL (d) and soleus (e) muscles from wild type and TRB3KO mice were dissected and incubated with tunicamycin (1 μg/ml) for 4 hrs (n=6-8). Data are the means ± S.E.M. * indicates p<0.05, ** indicates p<0.01, and *** indicates p<0.001 vs. basal in the same group. # indicates p<0.05, ## indicates p<0.01, and ### indicates p<0.001 vs. corresponding control, using One way ANOVA (a) and Two way ANOVA with Bonferroni correction.
