IL-23-mediated proliferation of stably transduced Ba/F3-gp130 cells involves PI3K/Akt activation. The indicated Ba/F3 cell lines were washed three times with PBS, starved for 4 h in serum-free medium, and pretreated for 60 min with P6 (1 μm), PD98059 (20 μm), or LY294002 (20 μm) before exposure to HIL-23 (0.2%) for 1 h. Cell lysates were analyzed for STAT3, Erk1/2, and Akt by Western blot analysis of total and phosphorylated STAT3, Erk1/2, and phosphorylated Akt using specific Abs. A representative example of two separate experiments is shown.
