Typical workflow of the click chemistry-based enzymatic assay. Alkyne lipids were used as substrates in enzymatic assays. After lipid extraction the alkyne moiety was quantitatively click-reacted with the fluorogenic dye 3-azido-7-hydroxycoumarin (symbolized as a star) to yield fluorescently labeled lipids. These were separated by TLC and analyzed by fluorescence detection.
