Effects of SeV infection on tetherin level and of tetherin suppression on VP production in HeLa cells. (a) HeLa cells were transfected with either scrambled (Scble) siRNA or a pool of tetherin-specific siRNAs (Teth) for 48 h before infection (m.o.i. = 20) with or without SeV (SeV +/−) for 24 h. Cell extracts were prepared and SeV N protein and actin levels were determined by Western blot analysis. Cell supernatants were collected and VPs were pelleted and quantified according to the N protein levels (VP). (b) Infectious particles in cell supernatants measured by flow cytometry and expressed as the number of infectious units (IU) ml−1.
