Sestd1 is a novel binding partner of Dact family members. A, Sestd1 interacts with all three Dact paralogs in HEK293T cells. HA-Sestd1 was expressed with or without FLAG-Dact1, FLAG-Dact2, and FLAG-Dact3 in HEK293T cells. Protein complexes were immunoprecipitated (IP) with anti-FLAG-agarose beads, and associated proteins were detected by immunoblot (IB) with anti-HA antibody. B–D, amino terminus of Dact1 interacts with the carboxyl terminus of Sestd1, including the third spectrin domain (Spec3). Appropriate FLAG-, HA-, or enhanced GFP-tagged proteins were recombinantly expressed in HEK293T cells. The co-IP assays were performed as in A.
