Schematic representation of the concept of eliminating replication competent wild-type–like AAV particles from a rAAV production system. Major components of the system proposed in this work and the traditional system are illustrated. The sequestration of the carrier DNA (‘rep&cap’) and rAAV-DNA into two different cellular compartments eliminates the physical contact of rep&cap and rAAV DNA and therefore the possibility of generating wild type (wtAAV), i.e. rcAAV particles. The ‘rep&cap’ denotes the corresponding genes and ‘Rep&Cap’ - proteins.
