PTH induces LRP6 aggregation and Gα_sβγ accumulation in UMR-106 cells, as analyzed by sucrose gradient sedimentation. UMR-106 cells were transfected with siRNAs specific for GFP or LRP6, and were treated with PTH(1–34) (50 nM) for the indicated time periods. Cell ly-sates were subjected to sucrose density-gradient ultracentrifugation, and fractions were analyzed by Western blotting with antibodies against LRP6, Gα_s, Gβ₁, and Gγ₂.
