CPT suppresses Drp1 and parkin mRNA expression. p53+/+ or p53−/− neurons were treated with DMSO (control) or CPT (5 μm) 3 d after plating for 12 h. Expression levels of Drp1 and parkin mRNA were analyzed by real-time RT-PCR (see Materials and Methods). The results obtained were first normalized against levels of ribosomal protein S18 (RPS18) mRNA used as an internal control for RNA loading and then presented relative to the levels in DMSO-treated p53+/+ neurons. The data are mean ± SEM three separate experiments done in duplicates.
