BB0172 localizes on the B. burgdorferi surface. B. burgdorferi B31A3 was cultured at RT/pH 7.6 (lane 1), shifted to 37°C/pH 6.8 (lane 2), and shifted to 37°C/pH 6.8 and treated with proteinase K (lane 3). Whole-cell lysates of all treatments were separated on a 12% SDS-PAGE gel and stained with Coomassie blue (A) or transferred to PVDF membranes and probed with mouse anti-BB0172pep followed by anti-mouse HRP-conjugated antibody (B). OspC (outer membrane lipoprotein), SodA (intracellular), and P66 (outer membrane integral protein) were used as controls for the proteinase K treatment. T/pH shift denotes the shift from RT/pH 7.6 to 37°C/pH 6.8.
