Bovine spermatozoa were incubated in capacitation medium for 4-7h. (A) Ca²⁺ ionophore A23187 (10 µM) was added for 20 min at the indicated times. (B) The cells were incubated in NKM medium (starvation) for 1h and then transferred to capacitation medium. Spermatozoa were incubated with or without calcium (2 mM) and BAPTA-AM (3 µM) for 4h. Proteins were extracted and analyzed by western blot using anti-AKAP3 antibody and anti-β-actin antibody (loading control). The results represent three independent experiments.
