Changes in the O-GlcNAcylation of cytosolic and nuclear NF-κB/Rel proteins induced by LPS with or without GlcN. BV2 cells were pretreated with GlcN (5 mM) for 2 h and then treated with LPS (0.1 μg·mL⁻¹) with or without 5 mM GlcN for 24 h. Cytosolic and nuclei lysates were prepared (Input) and each fraction was precipitated with WGA-conjugated agarose beads (Pull: WGA). The precipitates were immunoblotted by anti-p65, c-Rel, p50, p52, OGT or O-GlcNAc (CTD110.6) antibodies. The blots shown are representative of three independent experiments.
