Down-regulation of inflammatory genes induced by TX 527 and bortezomib. Stable SVEC-vGPCR cells targeted with small hairpin RNA against mouse VDR (vGPCR-shVDR) or control shRNA (vGPCR-shctrl) were cultured and treated with TX 527 (10 nM, 24 h) or vehicle (0.01% ethanol) for 24 h (A and B) or in the presence/absence of bortezomib (0.5 nM) (B). Total RNA was extracted and reverse transcribed (0.5–1 μg). Gene expression of IL-6, MCP and MIP3α was assessed by qRT-PCR analysis, normalized to β-actin RNA levels. The statistical significance of treated versus control (vehicle) conditions was evaluated using Student's t-test and is explained in detail in Results section. The data shown are representative of three independent experiments done in triplicate. Results were analysed by the two-tailed t-test. P-value and statistical significance are given in the Results section.
