Increased RBP-J reduces cytokine production. RAW 264.7 cells were transduced with retrovirus-containing vector only control or FLAG-tagged RBP-J as detailed under “Experimental Procedures.” Cells expressing high levels of RBP-J were selected and used for these experiments. After transduction and selection, these cells were exposed to 100 ng/ml LPS for 6 h. A–E, RT-qPCR was used to quantify transcript levels of Il6, TNF, Il10, Il1b, and Il12b. F–H, the levels of IL-6, TNF-α, and IL-10 secreted into the culture medium were measured by ELISA. IL-1β and IL-12 were not detected in the culture medium by ELISA.
