Primer extension reactions catalyzed by Klenow fragment (exo⁺). Using unmodified or N²-Me-dG-modified 38mer template (sequence 4 in Table 1) primed with a 5′-³²P-labeled 10mer (sequence 7 in Table 1) primer extension reactions were conducted at 20°C using 0.004 U Klenow fragment as described in Materials and Methods. One-third of the reaction mixture was subjected to denaturing 20% PAGE. 13× shows the location opposite the N²-Me-dG lesion.
