Figure 5.

Permeability measured using Bodipy Ouabain in type I–like RAECs that were stretched for 60 minutes at 37% ΔSA in comparison to unstretched (Uns) cells. The cells were treated with VC or with the NF-κB activation inhibitor MG132; the nitric oxide and superoxide scavengers cPTIO and Tiron, respectively; or with the extracellular signal–regulated kinase pathway blocker U0126. Data are expressed as mean ± SEM of fold change above unstretched cells treated with VC. *P < 0.05 versus unstretched VC; P < 0.05 versus stretched VC. Inset: Permeability measured using Bodipy Ouabain in unstretched and stretched (37% ΔSA for 60 min) type I–like RAECs with and without treatment with 2 mM 2-deoxy-d-glucose and 10 μM antimycin A to reduce the expression of tight junction proteins and to increase cell–cell spacing (6). Data expressed as mean ± SEM of fold change above unstretched cells treated with VC. *P < 0.05 versus unstretched VC; #P < 0.05 versus unstretched AMA. NS = no significant difference between the marked groups.
