mVps45 knockdown perturbs GLUT4 levels. (A) Whole-cell lysates of 3T3-L1 adipocytes infected with four different retroviruses harboring shRNA species designed to knock down mVps45, a random shRNA, or control (noninfected) cells immunoblotted with anti-mVps45 or GAPDH as a loading control. For each lane 50 μg of protein was loaded, and the position of molecular weight markers is shown. Data from a typical experiment are presented. (B) Whole-cell lysates from noninfected cells or cells infected with shRNA #1, shRNA #2, or random shRNA, immunoblotted with the antibodies indicated. Data from a representative experiment is shown; all were repeated a minimum of three times using different batches of cells infected with at least two different batches of the indicated virus. (C) Quantification of data of this type, in which the extent of knockdown is quantified. Data shown are mean ± SD with the data normalized against control cells. Asterisk indicates a statistically significant decrease in expression (see text for details; p ≤ 0.05 for each case).
