Deregulated expression of c-Myc elicits
apoptosis induction by cisplatin. Exponentially growing
Rat-1/MycERTM (myc) or Rat-1/ΔMycERTM
(Δmyc) cells were exposed to OHT for 16 h or left untreated
(lanes C in panel A, middle panels in B anc C) and then exposed to
cisplatin (cDDP) for 3 (C), or 6 h (A, B) at the indicated
concentration of 0, 10, 25, or 50 μM, or for 1 h at a
concentration of 25 μM (D). Lanes M contain DNA markers.
Internucleosomal DNA degradation (A), condensed and fragmented nuclei
(B), cell blebbing (C), and clonogenic survival (D) were evaluated as
described in MATERIALS AND METHODS. The means and SDs were calculated
from at least three separate experiments.
