Effects of dGTP concentration on telomerase activity and processivity. (A) Primer extension assays were performed using either TEL15 (lanes 1–3) or TEL15(m4,5) (lanes 4–6) and telomerase that was purified by DEAE chromatography. All reactions contained in addition to 0.2 µM of labeled dGTP and 50 µM of unlabeled dTTP, either 0 µM (lanes 1 and 4), 1 µM (lanes 2 and 5), or 3 µM (lanes 3 and 6) of unlabeled dGTP. The positions of the P + 3 and P + 6 products are indicated at both sides of the panel. A low molecular weight RNase-insensitive product (indicated by asterisks) can be observed in this set of assays. (B) The total levels of nucleotide incorporation for the TEL15 and TEL15(m4,5) primer are plotted at different dGTP concentrations. The dGTP concentrations (in µM) are indicated at the bottom.
