Selection of candidate siRNA sequences. (A) The starting rules for generating a siRNA database are shown. (B) Selection of five siRNAs for each of 30 mRNAs from genes whose protein products are within the PI3K pathway. siRNA selection was limited to the open reading frame (CDS) and the 3′UTR. Next, the CDS was divided into four equal quarters and the 3′UTR. In each region the first unique siRNA candidate within the database was selected. (C) siRNA sets were provided both as single candidates as well as in two pools that either contained (P1) or did not contain (P2) the position 5 duplex.
