K19 locus targeting strategy. (a) Structures of K19 alleles and targeting vector. The targeting vector has a 1.3-kb 5′-arm, which locates immediately upstream of the K19 ATG. The 3′-arm is a 7.2-kb fragment that has a 156-base pair overlap with the 5′-arm. The targeting vector uses Diphteria Toxin A (DTA) subunit for negative selection and a FRT-flanked neomycin resistant gene for positive selection. Pr1 and Pr2 indicate the oligos utilized for genotyping. (b) Southern blots to show several targeted ES cell clones that give expected hybridization patterns (see Materials and Methods section for the expected size of DNA fragments). Sp, SpeI, Ba.
