Slit2 does not impair neutrophil recruitment or bacterial clearance in a model of E. coli ascending urinary tract infection. N-mSlit2 (2 µg) or control vehicle was injected intravenously into mice 1 hour before intravesical inoculation with 109 CFU/ml (100 µl) uropathogenic E. coli CFT073 suspension; 6 and 24 hours later, urine was collected in a sterile fashion, and mice were euthanized. The kidneys were collected aseptically and homogenized, and 0.1 ml each homogenate were plated on tryptic soya agar. The number of colonies was scored after overnight culture at 37°C. (A) Urinary neutrophil concentration was calculated by hemocytometer analysis of fresh urine samples. Bacterial colony counts from (B) urine and (C) kidney homogenates are presented as log CFU per milliliter. Results were obtained from n=6 mice per group.
