UV/visible absorbance spectra of wild-type nitrate reductase and its K58Q and K130Q variants. Spectra were measured at ambient temperature in a 1.0 cm optical pathlength cell using solutions of either wild-type nitrate reductase or the indicated variant. All samples contained nitrate reductase at a concentration of 15 μM in 25 mM HEPES buffer (pH 8.0).
