Tumors from multiparous Casp2−/−/MMTV animals display a high mitotic index and abnormal cell cycle regulation. Tumors from multiparous (a) or nulliparous (b) Casp2+/+/MMTV (n=10 multiparous, n=11 nulliparous), Casp2+/−/MMTV (n=11 multiparous, n=11 nulliparous) and Casp2−/−/MMTV (n=20 multiparous, n=8 nulliparous) were examined and the mitotic index was calculated. Mitotic indices for each genotype are shown. Error bars represent the S.E.M. P-values were determined by one-way ANOVA. (cf) Casp2+/+ and Casp2−/− MEF were treated left untreated (c), or treated with 1 μℳ 5-Fluorouracil (d), 1 ng/ml nocodazole (e), or cultured in the absence of serum (f) for 16 h, at which point the media was replaced with fresh media and cells were cultured for an additional 24 h. Cells were harvested and the cell cycle was analyzed by propidium iodide staining. The percentage of cells in G1, S or G2/M phase for each cell line is shown.
