(A) Effect of increasing doses of doxorubicin on galectin-7 expression in MDA-MB-231, a mutated p53R280K cell line and MDA-MB-453, a p53null cell line. No increase in galectin-3 mRNA levels was observed in MDA-MB-453. GAPDH was used as loading control. (B) MDA-MB-231 were treated or not with 0.5 µg/ml of doxorubicin before cells fixation and permeabilization. A goat anti-human galectin-7 polyclonal antibody was used in combination with an Alexa Fluor 488 donkey anti-goat IgG to detect endogenous galectin-7. MDA-MB-231 cells are shown with galectin-7 (green) and nucleus (blue). (C) Inhibition of galectin-7 mRNA levels and p53 protein levels by p53 siRNA in MDA-MB-231 cells treated with 0.5 µg/ml of doxorubicin. A siRNA CTRL vector was used as control and GAPDH or β-actin was used as loading control.
