KLF4 KO DCs are relatively deficient in histone acetylation. The relative degrees of histone acetylation in the IL-6 promoter were analyzed by ChIP. Sheared chromatin was prepared from formaldehyde cross-linked BMDCs. DNA from chromatin complexes was immunoprecipitated with an antibody specific for histone H4K27ac then purified and analyzed by qPCR using primers specific for the IL-6 promoter.
