In comparison with wild-type PI5P4Kα control (50 ng), PI5P4Kβ mutant G1 showed enhanced activity over wild-type PI5P4Kβ (15 μg). Similarly, mutation of the PI5P4Kγ G-loop (G1–G3) enhanced activity over wild-type PI5P4Kγ (200 μg). Additional mutations further increased the activity of mutant PI5P4Kγ G3 (A, B and A+B, 20 μg of each). At least four replicates of each sample were used (n=15, 7, 7, 10, 4, 4, 4, 6, 6 and 9 respectively from left to right). For details of specific mutants see Figure 2.
