The effect of dimethyl sulfoxide (DMSO) on 14-3-3/Bad TR-FRET assay. (A) Increasing amounts of DMSO (0%–18%) were added to the TR-FRET reaction and incubated at RT for 2 h. The TR-FRET signals were measured, and the FRET signal windows were calculated and plotted against the amount of DMSO. (B) The quality of assay performance for HTS was evaluated by Z′ and S/B in the presence of increasing amounts of DMSO. Z′ and S/B were calculated as described in the Materials and Methods section.
