Figure 5. Membrane-LT expression on B cells is necessary for FDC activation.

Figure 5

Splenic B cells from CD45.2+, CD19+, mLT+ (CD19+/+, LTβflox/flox), CD19+, mLT (CD19+/cre, LTβflox/flox) or CD19, mLT (CD19cre/cre, LTβflox/flox) were adoptively transferred into CD45.1+, CD19−/− mice, after which the mice were immunized i.p. with SRBC. Ten days post-immunization, splenic FDC activation and GC formation were visualized with biotinylated PNA (blue), anti-CD35 mAb (red), anti-VCAM-1 mAb (green) or anti-CD16/32 mAb (green). CD45.2+ donor B cells were detected using anti-CD45.2 mAb (white). Overlay of VCAM-1 or CD16/32 with CD35 staining is visualized by yellow, whereas overlay of these markers in addition to PNA staining appears white. Scale bars are 100 μm. Images are representative of 3 independent experiments in which 3 mice per experimental group were immunized.
