The lipid raft-associated proteins CD29 and CD98 were not directly involved in VV binding to activated T cells. HeLa cells and primary human activated T cells were transfected with siRNA constructs against human CD29 and CD98, and knockdown was measured by Western blotting on HeLa cells (A) and surface expression on activated T cells (B). VV binding with vA5L-YFP and infection with VV-EGFP were performed on HeLa cells (C) and activated T cells (D) to analyze the effects on CD29 and CD98 on VV binding and infection. (E) A representative FACS plot of HeLa cell CD29 surface staining versus vA5L-YFP binding shows the relationship between CD29 expression and VV binding. NS, not significant; *, P < 0.05.
