N-demethylation of caffeine to xanthine by P. putida CBB5. NdmA catalyzes the N1-demethylation of caffeine to theobromine, which is then N3-demethylated to 7-methylxanthine by NdmB. Both NdmA and NdmB couple with NdmD for electron transfer from NADH. 7-Methylxanthine is N7-demethylated to xanthine by the NdmCDE protein complex. Each reaction requires NADH and oxygen, with the methyl groups removed as formaldehyde.
