Effect of LY294002 and U0126 on EGF-stimulated AKT and ERK1/2 activation of rat goblet cells. Cultured rat conjunctival goblet cells were serum starved for 24 hours. The cells were preincubated with the PI-3K inhibitor LY294002 (10−7–10−5 M) (A–C) or MEK inhibitor U0126 (10−7–10−5 M) (D–F) for 30 minutes followed by stimulation with EGF (10−7 M) for 5 minutes. Western blot analysis was performed using antibodies against phosphorylated and total AKT (A, B, D, E) and ERK (A, C, D, F).
