PPARγ is required for GLP-1 induction of Pdx1 expression in mouse islets. A, normal mouse islets were incubated 8 h with or without 100 nm GLP-1(7–36 amide) followed by real time PCR analysis. Expression of PPARγ and Pdx1 was doubled compared with no observable effect on MafA or Foxa2 expression. B and C, that protocol was used in isolated islets from 8-week-old floxed control mice (B) and PANC PPARγ^−/− mice (C) with the normal GLP-1 induction of Pdx1 expression absent in the PANC PPARγ^−/− islets.
