Splenocytes and lymph node cells were harvested from WT, IL-1R^−/−, or MYD88^−/− mice. (A) Sorted naïve CD4⁺ T cells (CD4⁺CD44^loCD25⁻) were cultured with WT bone marrow-derived dendritic cells (BMDCs) and stimulated with anti-CD3 alone or in the presence of IL-1β or combined IL-1β and IL-23. Supernatants were collected at 72 hrs and cytokines were measured by ELISA. (B) Sorted γδ T cells were stimulated with IL-1β, IL-1β and IL-23, or IL-1β and IL-12. Supernatants were collected after 72 hrs and cytokines were measured by ELISA. Data are representative of two independent experiments with 2–3 mice per group. Data represent mean ± SEM.
