OPG production by PASM cells and heterogeneous mixtures of cells grown from LAM lung. Serum-starved PASM cells and three different heterogeneous mixtures of cells grown from LAM lung from three different patients (100,000 cells per well of a 6-well plate) were incubated for 24 hours in serum-free medium (SF), with or without 100 ng/mL CCL2 or 100 ng/mL TNF-α, or complete medium (CM), and culture supernatants were collected for quantification of OPG concentrations by ELISA. PASM cells were tested six times, as was each of the heterogeneous mixtures of cells grown from LAM lung. LAM data are reported as a LAM cell average. Data for proliferation stimulated by complete medium or with TNF-α or CCL2 are percentage of serum-free medium control and were normalized to the final number of cells per well.
