The yeast wild-type (WT) and mutant strains transformed with or without complementation constructs were streaked onto minimal medium (MM) with 2% galactose as the carbon source and 10 mM urea or proline as the sole nitrogen source. The WT strain grew well on all media whereas the mutants survived only on 10 mM urea. The mutants were able to grow on MM supplemented with 10 mM proline when the native Put1 or Put2 of the yeast or C. parasitica Prodh or P5Cdh fused to the yeast Sdh1 mTP was re-introduced into the corresponding mutant.
