Ferret ASF inhibits rAAV2/1 virus uptake and transduction in HeLa cells. HeLa cells cultured on a 48-well plate (∼10⁵ cell/well) were infected with rAAV2/1.CMVluc preincubated with 50 μl ferret ASF or 50 μl PBS at an MOI of 10³ DRP/cell. Infections were performed for 4 hr in the absence or presence of 1 μM Dox, and then the inoculums were removed and the cells were replenished with fresh medium. (A, B) Quantification of internalized viral genomes (A) and luciferase transgene expression (B) in cell lysates were performed at 24 hr postinfection. (C) An index for rAAV2/1 vector efficiencies after the indicated infection conditions was calculated as the ratio of luciferase expression divided by the number of viral genome.
