Optimum pH of extracted alkaline protease. Optimum pH was obtained by incubating the enzyme with substrate at different pH values. The buffers used were 0.03 M phosphate (pH 6.0–8.0); 0.1 M Tris–HCl (pH 8.0–9.0); 0.1 M glycine–NaOH (pH 9.0–10.0) and 0.03M borax–NaOH (pH 9.5–11). The activity at pH 9.0 is found as optimum.
