MDM2-ALT1 and MDM4-ALT2 are expressed in human RMS tumor samples. (A) Total RNA was isolated and assayed for MDM2 and MDM4 splicing by RT-PCR in ARMS, ERMS, and anaplastic RMS samples as well as in matched normal muscle tissue of five patients with RMS. Occurrence of splice variants MDM2-ALT1 and MDM4-ALT2 is strongly associated with ARMS and ERMS subtypes but not with anaplastic RMS. (B) The statistical significance of the MDM2-ALT1 splicing between each group was determined using the two-sided Fisher exact test. There was a significant difference in expression of MDM2-ALT1 between the alveolar or embryonal subtypes compared to the anaplastic RMS subtype (P < .0001). (C) All the RMS tumor samples were analyzed for correlation of the presence of MDM4-ALT2 with subtypes of RMS as in B. There was a significant difference in expression of MDM4-ALT2 between the alveolar (P = .0007) or embryonal (P = .0017) subtypes compared to the anaplastic RMS subtype.
