Monopolin is required for proper metaphase sister centromere separation distance. (A) Control and csm1Δ/Δ TetO-CEN7/TetR-GFP cells were grown in SDC-glucose until cells reached log phase. Log-phase cells were adhered to concanavalin-coated glass coverslips and imaged in SDC media at 30°C. Distances between two TetR-GFP spots in metaphase (brackets) were then measured. Representative images are shown. Scale bar, 1 μm. (B) The distance between sister centromeres was determined by Gaussian fitting to the diffraction-limited TetR-GFP spots. Significance of centromere distance differences between wild-type and csm1Δ/Δ metaphase cells was determined using a Welch's two-sample, two-tailed t test conducted in R. *p << 0.001.
